An in vitro synthetic biosystem based on acetate for production of phloroglucinol

BACKGROUND Phloroglucinol is an important chemical, and the biosynthesis processes which can convert glucose to phloroglucinol have been established. However, due to approximate 80% of the glucose being transformed into undesirable by-products and biomass, this biosynthesis process only shows a low yield with the highest value of about 0.20 g/g. The industrial applications are usually hindered ...

Toward low-cost biomanufacturing through in vitro synthetic biology: bottom-up design

While most in vitro synthetic biology projects are usually used for the purposes of basic science research or the formation of high-value products, cell-free synthetic pathway biotransformation (SyPaB), which can implement complicated biochemical reactions by the in vitro assembly of numerous enzymes and coenzymes, would be used for low-cost biomanufacturing. In this article, we present bottom-...

A microbial biomanufacturing platform for natural and semi-synthetic opiates

Opiates and related molecules are medically essential, but their production via field cultivation of opium poppy Papaver somniferum leads to supply inefficiencies and insecurity. As an alternative production strategy, we developed baker's yeast Saccharomyces cerevisiae as a microbial host for the transformation of opiates. Yeast strains engineered to express heterologous genes from P. somniferu...

Biofuel production by in vitro synthetic enzymatic pathway biotransformation.

Cell-free synthetic pathway biotransformation (SyPaB) is the implementation of complicated biochemical reactions by in vitro assembling a number of enzymes or their complexes and coenzymes. Assembly of numerous enzymes without cellular membrane, gene regulation, or undesired pathway can circumvent some of the obstacles to modifying living microorganisms. Several synthetic pathways for the produ...

Immobilization of Cu(II)-Histidine Complex on Al-MCM-41 as Catalyst for Epoxidation of Alkenes

Cu(II) complex with L-histidine (His) ligand, immobilized within Al-MCM-41, designated as Cu(His)2/Al-MCM-41 was prepared and characterized by powder X-ray diffraction (XRD), nitrogen adsorption desorption, FTIR, UV-Vis and chemical analysis techniques. The surface area and pore volume were found to decrease after immobilization of Cu(II) complex on Al-MCM-41. It was found that Cu(His)2/Al-MCM-...
